Chalki to Agia Marina on Leros takes a little over three hours on the Dodekanisos Express, the Dodekanisos Seaways catamaran that works its way up the Dodecanese chain. It is a fast craft with airline-style seating rather than a big conventional ferry, and it calls at several islands en route, so the journey is broken up by short stops. Deck access is limited compared with a Blue Star ship.
Agia Marina is one of the two main harbours on Leros and the commercial and administrative centre of the island, with a working harbour feel and Italian-era architecture around it. The clocktower and the covered market by the waterfront date from the Italian administration between 1912 and 1943, and that period shapes much of what you see. Leros itself is a quieter island than Kos or Patmos, known for calm beaches and its wartime and Italian history.

Leros's other port, Lakki, handles the larger conventional ferries and is a few kilometres away, so it is worth checking both when planning onward travel.
