Mykonos to Mesta is a long conventional crossing of around two hours forty on the Blue Star Patmos, running north-east out of the Cyclades towards Chios. It is a seasonal working of the line that links the Cyclades with the North Aegean, so it appears in the timetable rarely rather than as a standing service. The Blue Star Patmos is a large ferry with lounges, deck space and vehicle capacity.
Mesta is a fortified medieval village in the mastic-producing south of Chios, known for its labyrinth of stone lanes built to keep out pirate raids. It has an atmospheric, inland feel despite sitting near the coast, with traditional tavernas and mastic heritage as the main draws. The area's principal ferry gateway is Chios Town, roughly 30km away, so onward transport is worth arranging.
