Blue Star Ferries run a small number of sailings from Piraeus to Mesta on the southwest coast of Chios, using the Blue Star Patmos. The crossing takes around seven hours, generally overnight, on a full-sized car ferry with cabins and deck seating. It is a useful direct approach to the southern end of Chios without going via the main town.
Mesta itself is a fortified medieval village in the mastic-producing Mastichochoria, and it is the reason most people come. The stone lanes were laid out defensively against pirate raids and still form a labyrinth, with the village feeling more inland than coastal despite the nearby harbour. Mastic heritage, traditional tavernas and the neighbouring villages of Pyrgi and Olympi fill an easy couple of days.
Sailings are infrequent, roughly weekly and seasonal, so build your plans around the timetable rather than the other way round. Current departures and fares are shown below, starting around forty euros. Chios town, the island's main port with far more frequent connections, is about thirty kilometres away by road.
