Mykonos to Karlovassi on Samos takes between about three hours forty and four and a half hours on the Blue Star Myconos, depending on the calls it makes at Ikaria and Fourni. It leaves from Tourlos, the newer Mykonos port north of town, and heads east across the Aegean. This is a conventional ferry with decks, lounges, a canteen and vehicle capacity.
Karlovassi is the second town and port of Samos, on the north coast west of the capital Vathy. It has more of a working-town character than the tourist-focused parts of the island, with a university presence and an old and a new quarter. Travellers use it as a gateway to the west of Samos and the beaches on that side, with buses and taxis running from the port.
